Biological Activity
(R)-WAC-224 is an anticancer quinolone taht demonstrates cell growth inhibition against venetoclax-resistant AML cells (MOLM-13, IC50=0.02 uM), directly inhibits topoisomerase-2 (Topo-II) and exhibits antileukemic effects both in vitro and in vivo.
(R)-WAC-224 reduced MCL-1 expression in parental and VEN-resistant AML cells
The combination of (R)-WAC-224 and venetoclax strongly induced cell apoptosis through DNA damage.
(R)-WAC-224 downregulated MCL-1 levels via the cleaved caspase-3 pathway in parental and venetoclax-resistant AML cells.
Azacitidine, venetoclax, and (R)-WAC 224 demonstrated antitumor effects against both parental MV4-11 and venetoclax-resistant MV4-11 cells in a xenograft mouse model.
