Biological Activity

19MS-5 is a specific small molecule CD28 inhibitor, exhibits direct CD28 binding (MST KD=12.48 uM) and potent inhibition of CD28-B7 interactions with IC50 of 5.2 uM in cellular reporter assays.
19MS-5 effectively inhibits CD28:B7-1 (CD80) interactions with IC50 of 7.83uM in ELISA-based inhibition assays.
19MS-5 inhibits T cell activation markers in tumor–PBMC co-culture,  suppresses CD28-dependent cyto-kine release, including IFN-γ, IL-2, and TNF-α, indicative of strong CD28-mediated T cell activation.
