6-Ethylthioinosine (6-ETI, NSC 39368, SRI 1215) is a nucleoside analog that effectively and selectively inhibits Primary effusion lymphoma (PEL) in vitro and in vivo, but not to other lymphoma cell lines.
6-Ethylthioinosine (6-ETI) has LC50 of 30-435 nM against KSHV+ PEL cell lines, and inhibits NF-κB in PELs at 10-50 uM.
6-Ethylthioinosine (6-ETI) requires adenosine kinase (ADK) to phosphorylate and activate 6-ETI.
6-Ethylthioinosine (6-ETI) induces ATP depletion and cell death accompanied by S phase arrest and DNA damage only in ADK-expressing cells.
6-Ethylthioinosine (6-ETI) shows a robust antitumor effect in a disseminated MM mouse model.
