Alintegimod (7HP349) is a small molecule activator of the integrins αLβ2 (LFA-1) and α4β1 (VLA-4). 7HP349 directly activates integrin cell adhesion receptors crucial for the generation of an immune response.
7HP349 increases tumor-specific T cell activation and localization to a non-T cell inflamed cold tumor microenvironment typically characterized by low ICAM-1 expression and enhances the anti-tumor activity of CTLA-4 and PD-1/L1.
7HP349 monotherapy had modest effects on anti- programmed death 1 (PD-1)-resistant tumors, whereas combinatorial treatment with anti- T-lymphocyte-associated protein 4 (CTLA-4) therapy increased CD8+ Teff intratumoral sequestration and synergized in inducing cancer regression, in cooperation with neutrophils.
