A16B1 is a specfic, allosteric inhibitor of the PhoQ histidine kinase, selectively inhibits the activity of the Salmonella PhoP/PhoQ system, blocks Salmonella pathogenicity.
A16B1 shows no activity against EnvZ/OmpR, CpxA/CpxR, PmrA/PmrB and Rcs TCS.
A16B1 inhibits the PhoP/PhoQ system by acting on the PhoQ domain that encompasses the second transmembrane region of the protein, negatively regulating its kinase activity.
A16B1 the antimicrobial peptide colistin shows a 4-fold reduction in its minimum inhibitory concentration (MIC) value when it is combined with A16B1.
A16B1 exhibits anti-virulence properties by decreasing Salmonella intramacrophage replication and increasing the survival of Galleria mellonella larvae, a complete multicellular organism model organism for the analysis of Salmonella systemic infections.
