Biological Activity

AAK1 inhibitor (S)-31 is a potent, selective and brain penetrant adaptor protein 2-associated kinase 1 (AAK1) inhibitor with IC50 of 5.8 nM.
AAK1 inhibitor (S)-31 displays 25-fold selectivity over BIKE (IC50 = 0.161 uM), and good kinase selectivity against in-house panel of kinases.
AAK1 inhibitor (S)-31 (30 mg/kg, sc) effectively reduced μ2 phosphorylation in C57BL6 mice.
AAK1 inhibitor (S)-31 exhibited improved enzyme and cellular potency compared to BMS-911172.
