ABD646 is a selective, covalent, small-molecule inhibitor of ABHD18, rescues TAFAZZIN (TAZ) mutant phenotypes in fibroblasts from human patients and in fish embryos.
ABD646 may bind to ABHD18(S199) covalently.
ABD646 shows a near-complete blockade of ABHD18 activity with modest selectivity at both 1 μM and 10 μM, as measured by the reduction of FP probe labelling.
ABD646 is also active in THP1 cells, as demonstrated by a roughly 80% inhibition of probe binding to ABHD18.
Chemical inhibition with ABD646 in cell culture matched the genetic inactivation of ABHD18.
ABD646 treatment (1 μM daily for 5 days) of zebrafish embryos largely restored heart morphology and function in Taz-deficient fish embryos.
