ACD855 (Ponazuril) is a potent, pan-Trk positive allosteric modulator (PAM) with EC50 of 1.9, 3.2 and 0.8 uM for TrkA, TrkB and TrkC respectively. ACD855 effectively potentiates the response of NGF and BDNF in TrkA- and TrkB-overexpressing cell lines . ACD855 facilitates LTP induction in hippocampal rat slices. ACD855 (3 or 10 mg/kg, s.c.) reverses memory-impairment in the passive avoidance task in mice.
