Biological Activity

AK3280 (GDC-3280, RG-6069) is an orally available, potential novel anti-fibrotic small molecule by inhibiting the ASK1-p38 MAPK signaling pathway.

AK3280 inhibits expression of fibrotic genes including Col1a1, Col1a2, Col3a1, and Acta2 induced by TGF-β, LPA, and serum in cell-based assays.

AK3280 exhibited anti-inflammatory and antifibrotic activity by inhibiting the ASK1-p38 MAPK signaling pathway in silicosis mouse model.

AK3280 attenuated the upregulation of fibrosis and TGF-β expression and improved cardiac function following Ang II stimulation.

AK3280 attenuated Ang II-induced cardiac fibrosis in mouse.
