Biological Activity
ALS-04 is a potent, dual inhibitor of TBK1 and ALK with IC50 of 378 nM and 9.5 nM respectively, effectively suppressing ALK-AKT signaling via the STING pathway.
ALS-04 inhibits cGAMP-induced immune response in THP-1 cells and Raw264.7 cells with IC50 of 1.62 μM and 1.97 μM, Imax of 95.7 % and 98.1 %, respectively.
ALS-04 inhibits cGAMP-induced type I IFN response.
ALS-04 inhibits cGAMP and LPS-induced inflammatory response.
ALS-04 significantly inhibits the secretion of both innate and pro-inflammatory cytokines in response to cGAMP and LPS stimulation in vivo.
