Biological Activity

ARVib-7 is a small molecule inhibitor of NDUFS4 that decreases NDUFS4 protein levels, a nuclear-encoded subunit of electron transport chain complex I, disrupts OxPhos and induces ferroptosis, ARVib-7 is a potent inhibitor of androgen receptor AR/AR variants, inhibits AR-V7 expression.

ARVib-7 (1 uM) inhibits AR by 91.64% and AR-V7 by 92.06% in CWR22Rv1 prostate cancer cells that express both AR and AR variants.

ARVib-7 (1 uM) inhibits AR by 93.59% and AR-V7 by 89.20% in CWR22Rv1 cells.

ARVib-7 (1.5 uM) disrupts the AR/AR-V7 gene expression program in enzalutamide-resistant prostate cancer cells C4-2B MDVR.

ARVib-7 (1 uM) inhibits AR/AR-V7 protein expression via HSP70/STUB1-mediated ubiquitin-proteasome system regulation in C4-2B MDVR cells.

ARVib-7 suppresses resistant prostate cancer cell growth and induces apoptosis.

ARVib-7 (25 mg/kg i.p) suppresseed advanced prostate tumor growth in mice bearing CWR22Rv1 xenografts

ARVib-7 (150 mg per kg p.o) suppresses advanced prostate tumor growth in mice bearing LuCaP 35CR xenografts.

ARVib-7 inhibits NDUFS4, disrupts mitochondrial function, and enhances olaparib sensitivity in resistant prostate cancer cells.
