Biological Activity
AS1670542 is a potent, selective, orally-active human thrombopoietin receptor (TPO receptor) agonist, concentration dependently promotes proliferation of Ba/F3 cells expressing human TPO receptor with EC50 of 1.9 nM.
AS1670542 promotes megakaryocyte colony formation from human cord blood CD34+ cells in a concentration-dependent fashion (EC50=260 nM).
AS1670542 induced tyrosine phosphorylation of STAT5 in human blood platelets, but not rhTPO induced tyrosine phosphorylation of STAT5 in beagle dogs, rats, and cynomolgus monkeys.
AS1670542 (0.3 and 1 mg/kg/day) significantly increased the number of human platelets on in vivo human platelet production in NOD/SCID mice transplanted with human fetal liver CD34+ cells.
