AT-121 hydrochloride (AT121) is a potent, selective bifunctional nociceptin/orphanin FQ peptide / Mu opioid peptide (NOP/MOP) receptor agonist with Ki of 3.67/16.49 nM, respectively.
AT-121 showshigh potency (EC50=35 nM) and partial agonist efficacy at the NOP receptor relative to N/OFQ, and high potency (EC50=20 nM) and partial agonist activity at the MOP receptor, relative to DAMGO.
AT-121 shows significantly lower binding affinity at the DOP or KOP receptors (Ki >100 nM).
AT-121 (0.003-0.03 mg/kg, s.c. injection) produces potent antinociceptive and antiallodynic effects in nonhuman primates and humans.
AT-121 does not induce side effects commonly associated with opioids, such as respiratory depression, abuse potential, opioid-induced hyperalgesia, and physical dependence.
