AZ Dyrk1B 33 is a potent dual-specificity tyrosine phosphorylation-regulated kinase (DYRK) inhibitor with IC50 of 16.8/39/254/430 nM for DYRK1A/1B/2/3, respectively, does not inhibit DYRK4 (IC50>30 uM).
AZ Dyrk1B 33 decreased glucose levels in both larval and juvenile fish, recapitulated the effect on Tg(gip:nLuc) zebrafish at a lower concentration, also increased the number of K- and L-cells, did not change the number of beta-cells.
AZ Dyrk1B 33 promoted enteroendocrine cell differentiation through Nfatc4.
AZ Dyrk1B 33 (10 mg/kg, oral gavage) increases the prevalence of incretin+ cells and improved glucose control in diabetic mice.
