Apicidin (OSI 2040) is a fungal metabolite that exhibits potent, broad spectrum antiprotozoal activity in vitro, potently inhibits HDAC with IC50 of 0.7 nM in an enzyme activity assay.
Apicidin induces morphological changes, cell cycle arrest at G1 phase, and accumulation of hyperacetylated histone H4 in Hela cells, changes the expression of p21WAF1/Cip1 and gelsolin.
Apicidin upregulates Foxa2 expression via hyperacetylation of H3 and a decrease in H3K27 trimethylation on the promoter regions.
