Astin C is a specific, cyclopeptide inhibitor of cGAS-STING signaling, binds to STING C-terminal binding domain (CBD) with Kd of 2.37 uM (STING-CTD-H232), blocks the recruitment of IRF3 onto the STING signalosome and restricts STING activation and signaling.
Astin C exhibits inhibitory effect on intracellular DNA-induced Ifnb expression is dose dependent, with IC50 (half maximal inhibitory concentration) values of 3.42 and 10.83 μM for mouse embryonic fibroblasts (MEFs) and IMR-90 cells (human fetal lung fibroblasts), respectively.
Astin C markedly attenuates the autoinflammatory responses in Trex1-/- BMDM cells and in Trex1-/- mouse autoimmune disease model.
