Autogramin-2 is a small molecule autophagy inhibitor with IC50 of 0.27 uM (amino acid starvation), specifically binds to the StART domain of GRAM domain containing 1A (GRAMD1A, Aster-A) and also specifically inhibits the cholesterol-transfer activity of GRAMD1A.

Autogramin-2 is a small molecule autophagy inhibitor with IC50 of 0.27 uM (amino acid starvation), specifically binds to the StART domain of GRAM domain containing 1A (GRAMD1A, Aster-A) and also specifically inhibits the cholesterol-transfer activity of GRAMD1A.
Autogramin-2 blocks cholesterol accumulation at autophagosomes and lysosomes in response to starvation in the presence of chloroquine.
Autogramin-2 does not compete with binding of 22-NBD-cholesterol to GRAMD1B (Aster-B) or GRAMD1C (Aster-C).
