Biological Activity

Avibactam sodium (NXL104) is a covalent and reversible non-β-lactam β-lactamase inhibitor with IC50 of 8 nM and 5 nM for β-lactamase TEM-1 and CTX-M-15, respectively, also is a specific inhibitor of ZDHHC3 with Kd of 1.09 uM (MST assay).

Avibactam sodium destabilizes SCAP, reduces SREBP2 transcription, and inhibits HCC growth.

Avibactam sodium inhibits class A and C β-lactamases, but not metallo types and Acinetobacter OXA carbapenemases.

Avibactam sodium also decreases the protein levels of ZDHHC3 and SCAP and liver total cholesterol levels, in a DEN/CCl4-induced mouse model of HCC.
