BC18630 (BC 18630) is a small molecule inhibitor of the E3 ubiquitin ligase subunit DCAF7, potently inhibits SARS-CoV-2 infection with IC50 of 39 nM in cell-based assays.
BC18630 decreases TFEB ubiquitination and significantly reduces the measured interaction between DCAF7 and TFEB in nucleus of cells.
BC18630 attenuates a wide range of viral infections including SARS-CoV-2 infection, prevents virally-triggered TFEB degradation and dose-dependently reduces OC43 coronavirus infection, reduces HCoV-OC43 NP expression with IC50 of 0.3 uM.
BC18630 (oral BID at 20mg/kg and 50 mg/kg for 5 days) dose-dependently reduced lung viral titer (PFU) in Syrian hamster model of SARS-CoV-2 infection, significantly reduced SARS-CoV-2 N-protein (NP) in lung, also reduced the observed level of lung injury, with the higher dose particularly reducing inflammatory infiltrates.
