BRD9 degrader ZZ7 is a specific, reversible covalent molecular glue degrader that selectively degrades BRD9, covalently engages DCAF16 at Cys178.
ZZ7 is a proteasomal pathway-dependent BRD9 molecular glue degrader.
ZZ7 recruits DCAF16 to induce the degradation of BRD9. induces selective degradation of BRD9 in synovial sarcoma cells, with minimal effect on human iPSC-derived cardiomyocytes.
ZZ7 shows antiproliferative activity synovial sarcoma cell lines HS-SY-II and 1273/99 with IC50 of 322 nM and 426 nM respectively.
ZZ7 induces tissue-specific BRD9 degradation, resulting in a potent and selective antiproliferative effect against synovial sarcoma cells while potentially avoiding the cardiac side effects associated with clinical-stage BRD9 degraders.
