Biological Activity
CAI0019 (CAI-0019) is a potent, selective bacterial carbonic anhydrase (CA) inhibitor with Ki of 66.9 nM and 346 nM for E. faecium α-CA and E. faecium γ-CA.
CAI0019 exhibits MICs ranging from 0.022 to 1.57 μM across the panel of 25 E. faecalis and E. faecium strains.
CAI0019 has wider range of MIC values of 0.094 to 50 μM for rarer Enterococcus strains.
CAI0019 exhibited a synergistic interaction with ampicillin and gentamicin.
CAI0019 (20 mg/kg PO, QD for 3 days) demonstrates potent antienterococcal activity in murine VRE peritonitis model after infection with E. faecium NR-31909.
