CARD9 inhibitor Compound 6 is a potent, specific CARD9 inhibitor with SPR Kd of 120 nM (human CARD9), and IC50 of 0.33 uM in TR-FRET assays, blocks CARD9-dependent NF-κB activation and cytokine release.
CARD9 inhibitor Compound 6 selectively suppressed the release of interleukin (IL)-1β, IL-6, and TNF via the Dectin-1/CARD9 axis in THP-1 cells, while TLR2/6-mediated cytokine secretion was unimpaired.
CARD9 inhibitor Compound 6 modifies cellular responses by acting directly at the level of CARD9 to prevent NF-κB signaling in primary human cells.
