Biological Activity

CCW 28-3 is a bifunctional BRD4 PROTAC degrader linking RNF4 ligand CCW16 to BET bromodomain family inhibitor JQ1, degrades BRD4 in a proteasome- and RNF4-dependent manner.
CCW 28-3 shows higher potency for RNF4 than CCW 16 with an IC50 of 0.54 μM in the competitive ABPP assay.
CCW 28-3 does not inhibit RNF4 autoubiquitination activity.
