    Biological Activity

    CD28 inhibitor 22VS is a selective small molecule CD28 inhibitor with IC50 of 7.8 uM in ELISA-based CD28:B7-1 (CD80) interaction assays, binds directly to CD28 with MST Kd of 52.45 uM.
    22VS effectively inhibits both CD28-CD80 and CD28-CD86 interactions in a dose-dependent manner, achieving IC50 values of 1.48 uM and 1.39 uM, respectively, in cell-based NanoBit luciferase assays.
    22VS specifically inhibits CD28-B7 interactions without inducing nonspecific cytotoxic effects.
    22VS inhibits CD28-B7 signaling in reporter- based blockade assay with IC50 of 6.96 uM.
    22VS (10-50 uM) demonstrates dose-dependent suppression of T cell activation markers (IFN-γ, IL-2, and sCD69), closely matching the response elicited by the biologic CD28 antagonist FR104.
    22VS (10-50 uM) suppresses CD28-dependent cytokine release in a human PBMC-mucosal co-culture model, suppresses CD28-induced cytokine production.
