CD28 inhibitor CA-23 is a potent, specific small molecule inhibitor of CD28, binds directly to CD28 with MST KD of 0.732 uM, inhibits CD28-B7 engagement and CD28-dependent T cell activation.
CA-23 exhibits no detectable binding to CD80, CD86, or CTLA-4 even at 100 uM.
CA-23 inhibits CD28-CD80 and CD28-CD86 interactions with IC50 values of 35.3 nM and 23.9 nM, respectively.
CA-23 inhibits B7 binding to cell-surface CD28, dose-dependently suppresses CD28-mediated T cell activation with an IC50 of 424.4 nM.
CA-23 blocks CD28-driven T cell activation in human blood without independent agonism.
CA-23 (1-10 mg/kg) ameliorates T cell transfer-induced colitis by suppressing pathogenic Th1 and Th17 responses in vivo.
CA-23 suppresses pathogenic cytokine and Th1/Th17 responses in PBMCs from patients with active IBD.
