Biological Activity
CRD-1441551 is a potent and highly selective, and reversible MALT1 inhibitor with IC50 of 33.5 nM.
CRD-1441551 shows 100-fold selectivity over 57 tested proteases tested at 3 uM.
CRD-1441551 decreased amounts of cBCL10 in LM-Y1, LM-Y2, and S1T cells, with IC50 values of 6.9, 10, and 18 nmol/L, respectively.
CRD-1441551 increased RelB expression in LM-Y2 and S1T cells.
CRD-1441551 inhibited the growth of ATL cell lines with GI50 of 20.4, 0.8, and 15.7 μmol/L in LM-Y1, LM-Y2, and S1T cells, respectively.
CRD-1441551 induced apoptosis in LM-Y2 cells, induced cleaved caspase-3 and -7.
CRD-1441551 (60 or 100 mg/kg) induced an increase in RelB protein expression and downregulated IRF4 protein expression in S1T xenograft model tumor tissue.
CRD-1441551 exhibited variable antitumor effects across ATL models both in vitro and in vivo.
