DC‐Rhoin (DC-RC-063) is a selective, covalent small molecule inhibitor of Rho family GTPase RhoA protein, inhibits GDP/GTP exchange rate of RhoA with IC50 of 2.94 uM.
DC‐Rhoin captures a novel allosteric pocket around cysteine 107 (Cys107), blocks the interaction RhoA with RhoGEF and RhoGDI.
DC‐Rhoin selectively inhibits the interactions between Rho family proteins (RhoA, Cdc42, and Rac1) and their respective GEFs.
Among the Ras superfamily of GTPases, the Cys107 residue is unique and relatively conserved within the Rho family.
