Biological Activity
Dual Hsp90/HDAC6 inhibitor 17 is a potent, selective dual Hsp90/HDAC6 inhibitor with IC50 of 28/96 nM respectively.
Compound 17 is 107- to 430-fold more potent on HDAC6 compared to class I HDACs and 18- to 96-fold more potent compared to class IIa HDACs.
Compound 17 is also 4 times more active on HDAC6 than HDAC11 and 29 times more active than HDAC10.
Compound 17 demonstrates cell growth inhibitory activity with GI50 of 0.7, 2.2 and 0.8 uM for LNCaP, PC3, and DU145 cancer cells.
Compound 17 demonstrated marked anticancer activity and ability to target both established tumor masses and tumor-initiating cell populations in 3D tumor spheroid models.
