Biological Activity
Elecoglipron (ECC5004/AZD5004) is a novel small‐molecule GLP‐1 receptor (GLP-1R) agonist, binds to the human GLP‐1R with IC50 of 2.4 nM, displays a full agonist profile with a potency of 2.1 nM.
not active against cell lines expressing mouse, rat and rabbit GLP‐1R.
ECC5004 actsas a partial agonist with an E max of 30% and an EC50 of 45 nM in CHO‐K1 cAMP assay.
ECC5004 does not demonstrate detectable β‐arrestin‐2 recruitment.
ECC5004 exhibits a dose‐dependent effect on potentiating insulin secretion at 11 mM glucose (EC50 = 5.9 nM) in EndoC‐βH5 cells.
ECC5004 reduces glucose and body weight.
