FOXO1-IN-10 (FOXO1 inhibitor 10) is a highly selective small molecule inhibitor of transcription factor FOXO1 with IC50 of 0.21 and 0.03 uM for hFOXO1 and mFOXO1, respectively.
FOXO1-IN-10 induces a robust cytotoxic response of mantle cell lymphoma (MCL) cells in vitro and suppresses MCL progression in vivo.
FOXO1-IN-10 is highly selective for FOXO1 both in vitro and in vivo and fails to elicit any effect in genetic models of FOXO1 ablation.
Chronic administration of FOXO1-IN-10 improved insulin sensitivity and glucose control in db/db mice without causing weight gain.
Chronic FOXO1-IN-10 treatment combined with FGF21 led to synergistic glucose lowering in lean, streptozotocin-induced diabetic mice.
