FSEN1 (Ferroptosis Sensitizer 1) is a specific small molecule, uncompetitive inhibitor of ferroptosis suppressor protein 1 (FSP1) with IC50 of 133 nM, sensitizes cancer cells to ferroptosis.
FSEN1 exhibits high potency in triggering cell death in the H460C GPX4KO cells with IC50 of 69.4 nM,
FSEN1 inhibits FSP1 CoQ1 oxidoreductase activity with IC50 of 688.3 nM, has no effect on the CoQ oxidoreductase activity of NQO1.
FSEN1 sensitizes multiple cancer cell lines of different tissue origins to ferroptosis, RSL3-induced cell death in a panel of cancer cell lines of various tissue origins, including lung (A549N), breast (HCC1143C), liver (Huh7C), glial (T98GN), bone (U 2-OSN), connective (HT-1080N), lymphoid (RL, SUDHL5), and skin (A375, SKMEL28,501-MEL).
FSEN1 is synthetic lethal with GPX4 inhibitors and sensitizes cancer cells to ferroptosis through inhibition of FSP1.
