Biological Activity
Ferritinophagy inhibitor 4k is a specific inhibitor of ferritinophagy, disrupt the interaction of NCOA4 and ferritin, inhibits RSL3-induces ferroptosis with IC50 of 520 nM in HT-1080 cells.
Ferritinophagy inhibitor 4k reduces cellular free Fe2+ and protect cells from ferroptosis.
Ferritinophagy inhibitor 4k binds to NCOA4 (Kd=0.78 uM, MST assay) and impairs NCOA4-ferritin interaction.
Ferritinophagy inhibitor 4k reduces cellular ferrous iron level and upregulates the ferritin protein expression.
Ferritinophagy inhibitor 4k could effectively alleviate ConA-induced acute liver injury in a mouse model.
