GANT61 (GANT-61) is a specific small molecule inhibitor of GLI1 and GLI2, inhibits Hedgehog (Hh) signaling pathway with IC50 of 5 uM in Gli reporter assays using murine NIH 3T3 cell line. GANT61 acts in the nucleus to block Gli function, inhibits both Gli1- and Gli2- mediated transcription, and demonstrates a high degree of selectivity for HH/Gli signaling. GANT61 inhibits Hh signaling downstream of Smo and Sufu. GANT61 inhibits GLI-dependent human tumor cell growth. GANT61 (50 mg/kg) blocks GLI function and subsequent cell growth, induces growth regression in human prostate cancer xenograft.
