GM47-1 is small molecule inhibitor targeting MmpL3 in Mycobacterium abscessus, has MIC50 of 1.6 uM and trigger a dose-dependent ATP increase with EC50 of 3 uM in Mycobacterium abscessus.
GM47-1 also acts as an uncoupler, leading to a decrease in intracellular ATP levels and a cytochrome bcc:aa3-dependent increase in oxygen consumption.
GM47-1 is potent against clinical isolates, bactericidal in vitro, and potent against intracellular Mycobacterium abscessus (Mabs).
GM47-1 blocks trehalose monomycolate (TMM) transport to the outer membrane, and Mabs growth susceptibility is mmpL3 dependent.
GM47-1 induces extracellular ATP release and uncouples respiration, potentiates the activity of β-lactam antibiotics.
