GPX4 inhibitor C18 (GPX4-IN-5) is a covalent inhibitor of glutathione peroxidase 4 (GPX4), shows remarkable inhibitory activity against TNBC cells with IC50 of 10-75 nM. C18 notably induces ferroptosis with high selectivity by increasing the accumulation of lipid peroxides (LPOs) in cells. C18 covalently binds to the Sec46 of GPX4. C18 exhibited tumor growth inhibition in the MDA-MB-231 xenograft model with a TGI value of 81.0% at 20 mg/kg without obvious toxicity.
