Biological Activity
HG043 is a potent thienopyrimidine-based GPR119 agonist with EC50 of 2.0 nM (human GPR119) in GPR119 CRE-bla CHO-K1 cells using a cAMP assay.
HG043 potently stimulates insulin secretion with an EC50 value of 0.1 nM from MIN6 mouse pancreatic β-cells, demonstrating approximately 100-fold higher potency compared to MBX-2982 (EC50 = 17.65 nM).
HG043 stimulates the secretion of GLP-1, Insulin, GIP, and PYY.
HG043 significantly enhances glycemic homeostasis in normal and diabetic rodents.
HG043 effectively reduces body weight, food intake, and fat mass in DIO rats.
