HMN-176 is an active metabolite of HMN-214 and inhibitor of mitosis, interferes with the subcellular spatial location of PLK1, does not inhibit PLK directly. HMN-176 inhibits meiotic spindle assembly in surf clam oocytes and delays satisfaction of the spindle assembly checkpoint in human somatic cells by inducing the formation of short and/or multipolar spindles. HMN-176 (2.5 μM) greatly increases the duration of mitosis in hTERT-RPE1 and CFPAC-1 cell lines. HMN-176 displays similar cytotoxicity against tumors with various characteristics from different organs.
