HZS60 is a specific NMDAR/TRPM4 interaction interface inhibitor, displays significant neuroprotective effects against both NMDA and oxygen-glucose deprivation/reoxygenation-induced ischemic injury in primary neurons.
HZS60 at both 10 μM and 100 μM showed no effect on calcium-activated TRPM4 channel activity.
HZS60 boosted NMDAR-mediated downstream signaling in responses to NMDA treatment in primary neurons, inhibited OGD/R-induced decrease in phosphorylation of ERK, CREB, CaMKII, and Akt levels
HZS60 eliminated NMDA-induced mitochondrial dysfunction in primary neurons.
HZS60 (ig, 25 mg/kg) reduced neuronal damage in the penumbra of cerebral brain of mice after I/R injury.
HZS60 (ig, 25 mg/kg) disrupted the N/T interaction in mice after ischemic stroke.
HZS60 (ig, 25 mg/kg) treatment inhibited microglial activation in the MCAO-treated mice.
