Biological Activity
IAP-08 is a potent, selective SMYD3 proteolysis targeting chimera (PROTAC) degrader with DC50 of 177 nM and Dmax of 90% in HiBiT-tagged SMYD3 expressing HN13 cells, effectively induces the formation of ternary complexes with SMYD3 and the XIAP E3 ligase.
IAP-08 significantly decreases the proliferative and invasive capacity of HPV-negative HNSCC cells.
IAP-08 (12.5 mg/kg, i.v.) shows a favorable pharmacokinetic profile in vivo.
IAP-08 regulates the transcription of cell cycle-, EMT- and immune-related genes in HPV-negative HNSCC cells.
