IBX-200 is a selective integrated stress response (ISR) modulator targeting stress sensor kinase GCN2, selectively eliminates ISR-high cells across diverse stressors.
IBX-200 inhibits opto-PKR cells with IC50 of 1.12 uM for 24 h in light, is not acutely cytotoxic to opto-PKR cells in dark uo to 50 uM.
IBX-200 increase ATF4 and CHOP expression, resulting in ATF4- and CHOP-mediated sensitivity to thapsigargin stress and ATP depletion as a hallmark of high ATF4 expression, upregulates ATF4 and DDIT3 (CHOP) in an ISR-pathway-specific manner.
IBX-200 selectively binds to GCN2 with Kd value of 25.2 uM.
IBX-200 selectively decreases GCN2 kinase activity with IC50 of 50 uM.
IBX-200 shows selective and broad-spectrum antiviral activity, decreases HSV-1 titers in vivo.
