Ibezapolstat (ACX-362E) is a potent, selective, first-in-class DNA polymerase IIIC (DNA pol IIIC) inhibitor with IC50 of 51 nM (B. Subtilis pol IIIC), inhibits C. difficile efficiently (MIC50, 2 ug/mL).
ACX-362E is a potent inhibitor of diverse clinical isolates of C. difficile with MIC50 of 2 ug/ml and MIC90 of 4 ug/ml.
ACX-362E shows preferential activity against C. difficile PolC over those of other organisms in vitro.
ACX-362E demonstrated therapeutic potential for the oral treatment of Clostridioides difficile infection (CDI).
