JWZ-8-103 is a potent TRIM21-targeting molecular glue triggering nuclear pore complex (NPC) degradation, is 10-fold more potent than PRLX-93936 against parental PANC-1 (IC50=9.5 nM) and H661 cells overexpressing high levels of ectopic TRIM21 (IC50 <10 nM).

JWZ-8-103 is a potent TRIM21-targeting molecular glue triggering nuclear pore complex (NPC) degradation, is 10-fold more potent than PRLX-93936 against parental PANC-1 (IC50=9.5 nM) and H661 cells overexpressing high levels of ectopic TRIM21 (IC50 <10 nM).
JWZ-8-103 promotes a robust and rapid interaction between TRIM21 and NUP98 (apparent EC50=∼20 μM for TRIM21 PRYSPRY and NUP98 APD).
PRLX-93936 reduces levels of other short-lived prosurvival proteins, including MDM2 and c-MYC, with cell line-specific depletion patterns observed across multiple cell lines (Hs766T, KP3, MIA-PaCa2, and TRIM21 OE H661 cells), further supporting the broader consequences of NPC degradation.
JWZ-8-103 (15 or 50 mg/kg, i.p. injection) is sufficient to deplete several NUPs, effective against PANC-1 cell line–derived tumor murine xenografts.
