JX3212 is a potent, specific and brain penetrant inhibitor of inwardly rectifying potassium channel 4.1 (Kir4.1, KCNJ10) with IC50 of 0.28 uM (−120 mV) and 0.19 uM (+50 mV).
JX3212 shows minimal inhibition against Kir channel subtypes (Kir1.1/2.1/2.2/4.2).
JX3212 shows effective antidepressant activity in the novelty‐suppressed feeding test (NSFT) and chronic social defeat stress (CSDS) model.
JX3212 demonstrates superior safety margins compared to both (S)‐ketamine and the conventional antidepressant imipramine in murine behavioral assays.
