Biological Activity
KCI838 is a specific small molecule inhibitor of androgen receptor (AR)-dependent growth by selectively binding to AR and blocking its association with ELK1, blocks AR binding to ELK1.
KCI838 strongly inhibited the BRET signal for binding of AR to ELK1, with 60% and 95% inhibition at 0.4 µM and 10 µM respectively in BRET assays.
KCI838 inhibits AR-dependent cell growth and colony formation in PCa model cells, KCI838 is inactive in AR-negative cell lines.
KCI838 selectively inhibited ELK1-dependent vs. androgen response element (ARE)-driven promoter and gene activation by AR.
