Biological Activity
KI-TOX-A3 is a specific TOX protein (thymocyte selection-associated high mobility group box) binder (KD=0.92 uM) and potent TOX-KAT7 PPI inhibitor with IC50 of 0.51 uM.
KI-TOX-A3 significantly reduced TOX-KAT7 interaction at 5 uM in Molt-4 cells overexpresses TOX protein.
KI-TOX-A3 could potentially destabilize the TOX protein in T-ALL cells, perturbed other potential TOX PPIs including KU70/80 and KAT7, modulates multiple interactions within the TOX interactome.
KI-TOX-A3 demonstrates potent and selective intracellular activity against TOX-dependent cell lines (T-ALL and CTCL cell lines).
KI-TOX-A3 impacts TOX protein levels, which may impact the TOX gene in transcription level with TF positive autoregulation.
