Biological Activity
LILRB4 inhibitor compound 4 is a high affinity, brain-penetrant leukocyte immunoglobulin-like receptor B4 (LILRB4, ILT3/CD85k) inhibitor, inhibits LILRB4-ApoE interaction with IC50 of 40.6 nM in ELISA assays.
LILRB4 inhibitor compound 4 suppresses LILRB4-dependent signaling, attenuates neuroinflammation, restores amyloid-β uptake, and shows no cytotoxicity in iPSC-derived human microglia.
LILRB4 inhibitor compound 4 exhibits high-affinity binding with MST Kd of 17.5 nM for ecombinant extracellular domain of mouse LILRB4.
LILRB4 inhibitor compound 4 (10 mg/kg, PO, QD) improved Y-maze performance and reduced regional Aβ42 levels, inflammatory cytokines, and CD86-positive microglia in male and female 5xFAD mice.
