LY3358966 is a potent, selective inhibitor of the intestinal sodium‐dependent phosphate cotransporter NPT2b (NaPi-2b, SLC34A2), inhibits phosphate uptake in human NPT2b expressed in CHO cells with IC50 of 32.4 nM.
LY3358966 also inhibits mouse and rat NPT2b expressed in CHO cells with similar potencies (IC50=43.9 and 26.8 nM, respectively).
LY3358966 also inhibits human NPT2a (IC50=11 nM), displays 9-fold selectivity over human NPT2c (IC50=251 nM), and no acitivity against human PiT1 and PiT2 (IC50>100,000 nM).
LY3358966 inhibits phosphate absorption in mice, effectively reduces acute phosphate uptake into plasma.
