Licochalcone D (Lico D) is a lycyrrhiza inflata-derived chalcone, potent and selective STING inhibitor, covalently modifies the Cys148 residue of STING and inhibits STING oligomerization. Licochalcone D (Lico D) significantly inhibits the degranulation in RBL-2H3 cells with low cytotoxicity. Licochalcone D (Lico D) significantly inhibited the Ca2+ influx and phosphorylation of extracellular signal regulated kinase (ERK) and MEK. Licochalcone D (Lico D) inhibits the recruitment of TBK1 and the nuclear translocation of IRF3 and NF-κB. Licochalcone D (Lico D) ameliorates the inflammatory phenotype in Trex1-1- mice and inhibits the progression of DSS-induced colitis and AOM/DSS-induced colitis-associated colon cancer (CAC).
