Lubiprostone (SPI-0211) is a potent, selective type 2 chloride channel (CLC-2) activator, increases electrogenic Cl- transport across the apical membrane of T84 cells with EC50 of 18 nM.
Lubiprostone (SPI-0211) activates ClC-2 Cl- currents in HEK293 cell line stably transfected with either recombinant human ClC-2 measured by whole cell patch clamp with EC50 of 17 nM.
Lubiprostone (SPI-0211) shows no effect on CFTR Cl- channel currents measured in CFTR-transfected HEK-293 cells.
Lubiprostone (SPI-0211) induces the passive movement of sodium and water into the intestinal lumen, yielding a net increase in isotonic fluid, induces intestinal secretion.
