Biological Activity

MA203 is a potent, selective Checkpoint kinase-1 (CHK1) PROTAC degarder with DC50 of 5 uM in MIA PaCa-2 (24 h).
MA203 (2 uM) highly significantly attenuated CHK1 levels by up to 80% and 50% in HU-treated MIA PaCa-2 and MOLT-4 cells, respectively.
MA203 not affect CHK2 and does not decrease GSPT1, also not alter IKZF1 or CK1α protein levels in MIA PaCa-2 and MOLT-4 cells.
MA203 induces CRBN-dependent proteasomal degradation mechanism.
MA203 promotes DNA damage upon HU-induced DNA replication stress.
